Identification of mutation-specific tumor infiltrating lymphocytes from metastatic ovarian cancer

چکیده
Introduction Novel interventions are needed for the treatment of women with metastatic ovarian cancer that causes 14,000 deaths each year in the USA. Retrospective analysis of tumor infiltrating lymphocytes (TIL) infused into patients who achieved complete, durable regressions of metastatic melanoma revealed that the responses were likely mediated through recognition of somatic non-synonymous mutations. Furthermore, prospective administration of mutation-specific TIL resulted in an ongoing partial response in a patient with cholangiocarcinoma.

Adoptively transferred tumor-infiltrating T cells target somatic cancer mutations in a human papillomavirus+ cancer patient with complete tumor regression
Background Adoptive transfer of tumor-infiltrating lymphocytes (TIL) can mediate complete regression of metastatic cervical cancer, but the immunological landscape of the anti-tumor T cell responses in these patients is not fully understood. Reactivity against the human papillomavirus (HPV)-derived antigens may contribute to the clinical responses, but whether other immunogenic tumor antigens a...

BACKGROUND/AIMS T-bet, a member of the T-box family of transcription factors, is a key marker of type I immune response within the tumor microenvironment, and has been previously reported by us to serve as an important prognostic indicator for human gastric cancer patients and a potential biomarker for immunotherapy. In the present study, we aimed to assess the clinical significance and prognos...
متن کاملDigital genomic quantification of tumor-infiltrating lymphocytes.
Infiltrating T lymphocytes are frequently found in malignant tumors and are suggestive of a host cancer immune response. Multiple independent studies have documented that the presence and quantity of tumor-infiltrating lymphocytes (TILs) are strongly correlated with increased survival. However, because of methodological factors, the exact effect of TILs on prognosis has remained enigmatic, and ...
متن کاملHigh-throughput characterization of the ovarian cancer HLA peptidome
Background T cells recognize cancer cells via human leukocyte antigen (HLA)/peptide complexes, and class I HLA-restricted tumor infiltrating T lymphocytes correspond to favorable prognosis in several cancers, including ovarian cancer–the most deadly gynecologic disease [1]. The accurate identification of tumor-specific HLA/peptide complexes is pivotal to their exploitation as targets in immunot...
متن کاملHER2/neu-derived peptides are shared antigens among human non-small cell lung cancer and ovarian cancer.
Previously, we have reported a correlation between the expression of HER2/neu and sensitivity to HLA-A2-restricted cytotoxic T-cells (CTL) in ovarian cancer. To investigate the role of HER2/neu in human non-small cell lung cancer (NSCLC), we established autologous tumor-specific CTL from tumor-infiltrating lymphocytes of HLA-A2+ HER2/neu+ NSCLC patients. These CTL lines specifically recognized ...
